I2C/GPIO was only tested on Linux.

The links you posted are for developing drivers. NUC5i5MYBE should already have I2C and GPIO driver support when they are enabled in the bios. When both are enabled in the bios the I2C controller is shown in Windows 10 Enterprise device manager as BhoomsC shows above , however, no GPIO controller is shown and the Windows 10 i2testtool from MS (Windows IoT - I2cTestTool) shows no i2ccontrollers found.
Is this solution header local IO not supported by NUC5i5MYBE for Windows 10 Enterprise?
As you know this thread is regarding Windows 10 Enterprise not Linux. The NUC5i5MYBE does have some Windows 10 Enterprise support via the Serial IO driver (Download Serial IO: Intel® Serial IO host controller for NUC5i[x]MY) for I2C. The GPIO controller is missing from the DeviceManager -- why? The I2C controller exists but where is the documentation to utilize it?
The specified Download Serial IO: Intel® Serial IO host controller for NUC5i[x]MY driver is not supported in Windows 10 Enterprise. After looking inside the installation log file I can say that it is Windows 8.1 specific. Can you provide more input on these?
