You can certainly develop your own programs on the NUC. In fact, for personal development projects, you can download and use, without charge, Microsoft's Visual Studio Community 2015 package. It supports the development of programs in C, C++, C#, Visual Basic, Javascript, Python and more. See here for more information (and downloads): https://www.visualstudio.com.
The NUC is not outfitted with an exposed RS232c-style serial port. Some NUC models have an internal expansion connector that allows you do gain access to, for example, serial bit buses like i2c and SMBus. Be aware, however, that the presence and features of the expansion connectors differ from one NUC model to another.
