This document: http://downloadmirror.intel.com/16959/eng/DQ45EK_SpecUpdate08.pdf says, that it is possible to install 8GB of RAM onto a Intel DQ45EK. So I took this RAM: http://www.mushkin.com/Memory/Silverline/996762.aspx (2x4GB) and installed it onto my board. Before I had 2x2GB.
This is the effect:
2x2GB installed: everything works fine
2GB in Slot A and 4GB in Slot B: works fine, too
4GB in Slot A and 2GB in Slot B: Management Engine tells me, that it failed to start, but system works.
4GB in Slot A and 4GB in Slot B: System hangs before BIOS. All HDDs are starting but there is no beep-sound and it doesn't show anything on screen and doesn't do anything.
4GB in Slot A and nothing in Slot B: same as 2x4GB
Nothing in Slot A and 4GB in Slot B: Management engine says, that there has been a memory decrease and there is nothing installed in Slot A
Deactivating the Management Engine doesn't change anything.
Changing the 4GB-modules doesn't change anything, too. Both seem to work, this doesn't seem to be the problem.
So how can I use 8GB of memory?

Updating drivers can fix problems even if the newer ones do not address the issue. A newer INF might kick the system into working. Could the discrepancy have to do with DDR667 being used for the 2GB modules? Such swaps can be disruptive but should work after restarts and/or updates. What BIOS version is present (I did see that is not mentioned in your Intel link)?

Old RAM was DDR2-800, too. So no change here. Old RAM was 2x2GB DDR2-800 5-5-5-16, new one is 2x4GB DDR2-800 5-5-5-18.
Updating the drivers? The board doesn't load the BIOS, so starting the system is impossible...
BIOS ist the newest version avaiable. Has to be CBQ4510H.86A Version 0127.

I sent this RAM back and bought new RAM. It is Mushkin/Hynix 2x4GB DDR2-800 CL6 and this works without any problems!

FYI, 4GB modules aren't listed as a supported configuration for this board:

It must have been the brand/model of RAM since Intel still lists 4GB modules and 5-5-5 for the board. The most stable system I have has 4x2GB 6-6-6-18 DDR2 800, with Win 7, virtualization active and 9600GT graphics. The power management of the DQ45 and DG45 series seems to differ from older systems. Sleep and hibernation should work better for these systems but add-ins like TV-tuner (particularly) or wireless and some newer USB keyboards have negative effects, at least with 2x2GB 800 at 6-6-6-18. My system with 2X1GB 667 + 2X1GB 800 at 5-5-5-15 Vista x64 has a monitor sleep, but not computer sleep/hibernation, issue I have been attributing to the old power supply or need for reinstallation.

Alas. Intel updated their "Supported Memory Configurations" in that PDF, but didn't do so on the website.
